Inbred Dark Agouti (DA) rats were originally obtained from the Zentralinstitut für Versuchstierzucht (Hannover, Germany) and Piebald Virol Glaxo(PVG) rats from Harlan UK Limited (Blackthorn, UK). The congenic line was established from DA and MHC-identical PVG.A rats using a speed-congenic approach with marker assisted selection. DA females were mated with male offspring selected from F2 (DAxPVG.A) with heterozygote alleles within chromosome 4 and Hse1 QTL interval and against lower PVG.A background contamination using 86 microsatellite markers equally spaced throughout the genome (at 20 centimorgan cM intervals). A breeding pair selected from the N5 generation were crossed for two generations to produce homozygous DA.PVGc4-Hse1-R1 congenic rats, carrying PVG alleles between D4Kini3 at 27.81 Mb and D4Mgh14 at 36.5 Mb. |
