Gene Affected Bscl2
Origin T239A mutation was found by screening of 4608DNA sample from ENU mutagenesis library KURMA. This strain has a T239A (L80X) nonsense mutation in Bscl2 gene.
Strain characteristics Bscl2T239A homozygous mutated rat shows severe atrophy of adipose tissue, hypoleptinemia, abnormal glucose tolerance, increment of triglyceride levels in the liver. Bscl2T239A mutated male rats have small testes with sperm hypoplasia and are infertile.
Breeding Conditions crossing between heterozygous rats
Genotyping Primers: Forward 5’-TGTGGGCCCAGGAAGTG-3’, Reverse 5’-CCCCAAACTGCAGCATCAG-3’ TaqMan MGB probes: WT probe 5’-CCTGCCAACACATGG-3’, mutant probe 5’-CCTGCCTACACATGG-3’ PCR condition: 95C 20 sec, 20 cycles od 95C 3 sec and 60C 20 sec
