Gene Affected |
tTA: tetracycline transactivator, Pvalb: Parvalbumin |
Origin |
background strain:Long-Evans rat(Iar:Long-Evans)
Transgene:
(1) Parvalbumin (Pvalb) promoter:mouse derived (MGI:97821). PV is calcium-binding protein.
(2) tetracycline transactivator (tTA):Fusion protein of tetracycline repressor (Escherichia coli-derived) and VP16 (Herpes simplex virus-derived). In the absence of Doxycycline (Dox), tTA binds to tetracycline-responsive element (TRE) and expression of TRE-controlled genes can be induced.
Vector:pBlueScript (Stratagene), pBACe3.6 (CHORI)
|
Strain characteristics |
tetracycline transactivator(tTA) is expressed in Parvalbumin-positive cells (neurons). In the absence of Dox, tTA binds to TRE and expression of TRE-controlled genes can be induced only in the Parvalbumin-positive cells.
Expression of TRE-controlled genes can be repressed by Dox. (Tet-Off system) |
Breeding Conditions |
Maintained by mating between heterozygous rats. |
Genotyping |
