Origin Mutant human amyloid beta (APP*) combined Thy1 promoter which is specifically expressed in neurons, was induced to ES cells from DA strain rats, and chimera rats obtained from the ES cells. The F1 rats were backcrossed with Crl:CD (SD) strain rats and F3 rats was deposited.
Strain characteristics The tendency of dementia-like behavior (declining object perception) is observed in a novel object recognition test. The expression levels of mutant human APP mRNA are difficult among Lines (12>1>14>6).
Breeding Conditions Mating the transgenic rats with wild type rats and maintaining as heterozygous rats.
Genotyping Decide the transgenic rat by PCR using the following primers. If approximately 9 kb fragments are amplified from the rat genome, the rat is decided as transgenic rat. Primer: TA-F 5’-GAATTCAGAGACCGGGAACCAAACTAG-3’ Primer: TA-R 5’- GAGAGAGAATAAGGACAGTGAACGAGTGG-3’
