Origin By CRISPR/Cas9 system, mutation was introduced in Cspg4 gene of Wistar-Imamichi rat (Iar: Wistar-Imamichi, Institute for Animal Reproduction). 7 bp deletion on Exon1.

Genotyping Run PCR at 2 times to distinguish between wild type and Cspg4 knockout (KO) rat.
Primer F: CTTAGAGACATTGCCCCAAATCTT Primer R: GATACCCGGAGTTCTCATAAACAGA
1st PCR: (DNA polymerase: KOD FX Neo) 94℃ 5 min, x 1 cycle 98℃ 10 sec, 58℃ 30 sec, 68℃ 40 sec x 38 cycles.
In the case of wild type or Cspg4/i> KO (homo), one DNA band is observed around 600 bp. In the case of Cspg4/i> KO (hetero), two DNA bands are observed around 600 bp.
2nd PCR : To distinguish between wild type and Cspg4 KO (homo), add the same amount of wild type rat genome into all PCR samples, and run PCR under the same conditions. In the case of wild type, one DNA band is observed around 600 bp. In the case of Cspg4/i> KO (homo), two DNA bands are observed around 600 bp.
