Gene Affected コンドロイチン硫酸プロテオグリカン4(CSPG4)
Origin By CRISPR/Cas9 system, mutation was introduced in Cspg4 gene of Wistar-Imamichi rat (Iar: Wistar-Imamichi, Institute for Animal Reproduction). 2 bp insertion on Exon1.
Strain characteristics Analyzing
Breeding Conditions Natural mating
Genotyping To distinguish between wild type and Cspg4 knockout (KO) rat, run PCR followed by treat the DNA fragments by using the restriction enzyme.
Primer F: CTTAGAGACATTGCCCCAAATCTT Primer R: GATACCCGGAGTTCTCATAAACAGA
PCR(DNA polymerasae: KOD FX Neo) 94℃ 5 min, x 1 cycle 98℃ 10 sec, 58℃ 30 sec, 68℃ 40 sec x 38 cycles
Cut DNA fragments by Mwo1 (restriction enzyme). Wild type: one DNA band is observed around 340 bp. Cspg4 KO (hetero): two DNA bands are observed around 200 bp and 340 bp. Cspg4 KO (homo): one DNA band is observed around 200 bp.
