This transgenic rat for labeling GRP-expressing neuron under the rat gastrin-releasing peptide (GRP)-promoter. In the cell having X-zone of lumbar spinal cord L3-L4 revel, 99% of GRP-expressing neuron are expressed Venus. |
Breeding Conditions |
Mate the heterozygous Tg rat and wild type Wister (Crlj:WI) and distinguish transgenic rats by genotyping the pups. |
Genotyping |
Amplify Venus gene by PCR. Venus primer sequence (forward): 5’-CACCATCTTCTTCAAGGACGAC-3’ (reverse) 5’-ATGATATAGACGTTGTGGCTGTTGT-3’PCR condition 95℃ 15 s, 50℃ 15 s, 72℃ 15 s 35 cycles, 72℃ 5min, Electrophorese the PCR products and distinguish Tg by check the band at 171 bp. |
