| Origin |
The purinergic receptor P2X4 (P2rx4)-deficient rat line was established at the National Cerebral and Cardiovascular Center Laboratory using the CRISPR/Cas9 system. SD rat (slc:SD) is a background strain. In line No. 18, conventional sequencing revealed genome deletion from the promoter region to the middle portion of exon 1 (171 b.p.) . |
| Strain characteristics |
No particular abnormalities are observed under normal rearing conditions.
The purinergic receptor P2X4 binds ATP and functions as a channel that allows Ca2+ to flow into the cell, and also functions as a shear stress sensor in vascular endothelial cells. |
| Breeding Conditions |
Breeding is performed by mating between homozygous deficiencies is possible. |
| Genotyping |
Determined by PCR.
KOD One PCR Master Mix (#KMM-101, Toyobo CO., LTD., Osaka, Japan) was used.
Primer sets; forward 5'-CGAGCTGACCCTGCTCCCTGG-3', reverse 5'-CCCTGCGCAGTCCCTTCACC-3'
Size; wild type; 732 b.p. mutant; 561 b.p. |
