Gene Affected Lepr: leptin receptor
Origin A congenic rat in which the leptin receptor gene Lepr on chromosome 5 derived from Zucker fatty rats was introduced by backcrossing for 10 generations into the genetic background of Wistar Kyoto rats (specific strain name unknown). By introducing the Lepr gene of Zucker fatty rats, which develop obesity but have normal blood glucose levels, into the genetic background of Wistar Kyoto rats, which have poorer glucose tolerance, the rats developed both obesity and diabetes. It was created by Ikeda et al. of Takeda Pharmaceutical Co., Ltd. and reported in 1981.

Breeding Conditions Since Lepr homozygotes are sterile in both sexes, mating is required between male and female heterozygotes.
Genotyping The Lepr genotype can be identified by PCR-RFLP using the restriction enzyme MspI. Primer-F: 5'-TATGGAAGTCACAGATGATGG-3', Primer-R: 5'-CTTACGATTGTAGAATTCTCTAA-3'.
