Origin This strain was generated by the group of Dr. Tomoji Mashimo, Institute of Experimental Animal Sciences, Graduate School of Medicine, Osaka University in 2017. Genome editing technology, the CRISPR/Cas9 system was used on LEW/CrlCrlj rats to replace five amino acids in exon 1 with human sequences (V53L, S62T, N64K, T120A, T149M) by homologous recombination using 1086 bp LssDNA. The founder was backcrossed to LEW/CrlCrlj rats once, and then homozygous rats were obtained by crossing between heterozygotes.
Strain characteristics No significant phenotype is observed macroscopically.
Breeding Conditions Maintained by mating of both homozygous rats.
Genotyping Based on the difference in sequence, the WT allele and humanized allele are amplified by PCR. WT sense: 5'-CATTAACTGGGGTGGCTCAGAGAACCCCCTACCTG-3' WT antisense: 5'-GCACAGAAATAAGAACAGAAGAGCTCTCACCGTG-3' humanize sense: 5'-CACCATTAACTGGGGTGGCACAGAGAAGCCTTTGC-3' humanize antisense: 5'-CAGCTCCGATGATGGCCCCGAGGCACTGCGCTGCG-3'
