This strain was created in collaboration with Professor Tomoji Mashimo of the University of Tokyo.
Genome editing was performed using CRISPR/Cas9 on fertilized eggs of SD rats, targeting exon 11 of the Brca2 gene, to introduce a frameshift with a stop codon. The genetic background is Jcl:SD. |
| Strain characteristics |
Brca2 exon 11 contains a frameshift/stop codon in the seventh BRC repeat.
Specifically, Brca2 p.T1942fs is homologous to human BRCA2 p.T1974fs.
Homozygotes for this mutation are embryonic lethal.
Sperm DNA fragmentation is observed in heterozygotes even at a young age. Furthermore, a decline in reproductive efficiency is seen in both males and females around eight months of age. |
