Origin BUF/Mna rats were established from sib mating of Buffalo rat colony transferred from Biological Associates in America to Department of Pathology, Nagoya City University in 1964. At F18 generation, the rats showed thymoma 100% and were named as BUF/Mna in 1977 (Matsuyama, 1977). Buffalo rat. (Oct 12, 2010)
Strain characteristics BUF/Mna rats develop spontaneous thymoma, muscle atrophy and nephropathy.
BUN/Mna starts to develop thymoma at 9 months of age (Matsuyama, 1988). The thymoma was defined as epithelial cell tumors by transplant experiments using ACI rats (Taguchi, 1992). A linkage study revealed that thymoma susceptibility was determined by an autosomal dominant locus Tsr1 (thymoma susceptible locus of rat 1) and it was located between D7Wox16 and D7Rat21 on chromosome 7 (Oyabu, 1999; Higo, 2000).
At pre-thymoma age, BUF/Mna rats have extremely large thymuses mainly in cortex. A linkage analysis showed two polymeric autosomal loci, Ten1 and Ten2, regulating thymus enlargement in BUF/Mna and they were mapped on chromosome 1 and 13, respectively (Murakumo, 1996; Ye, 2000).
BUF rats were susceptible to thymic lymphomas (Ogui, 1989) but resistant to MNNG-induced stomach carcinogenesis (Ohgaki, 1983). BUF rats spontaneously develop progressing atrophy of fast-twitch type muscle (Kato, 1982) and proteinuria associated with focal and segmental glomerulosclerosis (Nakamura, 1986). For those phenotypes, one or more QTL genes were mapped (Ogui, 1995; Amo, 1997; Murayama, 1998; Ushijima, 2000). (Oct 12, 2010)
