Origin A transgenic construct was designed with the rat prolactin promoter (-3221 - 3233) controlling EGFP. Transgenic rats originated from Crlj:WI (Wistar) rats. (Jul 16, 2009)
Strain characteristics In adenohypophysial cells, 55% cells are positive for GFP. Among GFP positive cells, 76% are immunostained with prolactin antibody. (Jul 16, 2009)

Genotyping 1. PCR analysis on genomic DNA. Primer sequences: Forward: 5'-CTGGCCACTATGTCTTCCTGA-3', Reverse: 5'-TGTAGTTGCCGTCGTCCTTGA-3'. Protocols: 95˚C-10 min, 35 cylcles of (94˚C-1 min, 64˚C-1 min, 72˚C-1 min) Product: about 860-bp. 2. Southern blotting with genomic DNA. EGFP + poly(A) (about 1-kb) is detected with EGFP cDNA probe on BamHI-cut genomic DNA. (Jul 16, 2009)
