Genotyping |
1. PCR analysis on genomic DNA. Primer sequences: Forward: 5'-CTGGCCACTATGTCTTCCTGA-3', Reverse: 5'-TGTAGTTGCCGTCGTCCTTGA-3'. Protocols: 95˚C-10 min, 35 cylcles of (94˚C-1 min, 64˚C-1 min, 72˚C-1 min) Product: about 860-bp. 2. Southern blotting with genomic DNA. EGFP + poly(A) (about 1-kb) is detected with EGFP cDNA probe on BamHI-cut genomic DNA. (Jul 16, 2009) |
