2.由来
クローズドコロニーであるDonryuラット(日本ラット、浦和)を発がん物質である3'-メチル-4-ジメチルアミノアゾベンゼン(3'-Me-DAB)を含有する飼料で飼育し、兄妹交配を20代以上繰り返しつつ発がんラットを淘汰することにより得られた[東胤昭氏(元阪大、バイオメディカルセンター)]。現在、普通飼料でも発がん耐性は固定され、近交系であることを確認、セアック吉富(福岡)で、SPFグレードでDRH/Seaとして維持・市販されている。
3.表現型・病態
3'-Me-DABを0.06%含有する飼料で、親株のDonryuラットを20週間飼育するとほぼ100%肝発がんが見られるが、同条件でDRH/Seaラットでは3%(1/33)にしか肝がんは認められなかった。DRH/Seaの発育・繁殖に関してはDonryuラットと同等である。Donryuラットでは、52週令で子宮内膜腫が発生するものが若干存在するが、DRH/Seaでは3'-Me-DABの存在下で選抜されたにもかかわらず52
週令では、肝、肺、腎、子宮などに自然発生腫瘍は全く認められなかった。3'-Me-DAB以外の3'-CH2OH-DAB、DAB、2-AAFあるいはDMNによる肝がん発生にも顕著な耐性を示す他、DMBAによる乳がん発生に対しても耐性であった。
肝における3'-Me-DABの代謝的活性化はDonryuより低く、解毒代謝酵素はDRH/Seaの方が高い活性を示したが、3'-Me-DAB-DNA付加体に関しては、経時的変化を含め、両者の間には顕著な差は認められない。3'-Me-DAB投与時に前癌病変と見なされるGST-P陽性
fociは、両ラットで認められるが、DRH/Seaラット肝におけるGST-P陽性
fociの拡大 (clonal expansion)は著明に抑制されていた。3'-Me-DAB
による細胞毒性に関してもDRH/Seaは抵抗性を示す。
4.病因(原因遺伝子)
F344と DRH/Seaとを交配したF1ラットでは
3'-Me-DAB投与による肝RNAのGST-P
mRNAの誘導は親株のF344ラットと比較して顕著に抑制される。さらにF344 X
DRHF1ラットを兄妹交配し得たF2ラット108匹を3'-Me-DAB処理8週間後に、肝のGST-P陽性fociの数、平均サイズ、GST-P陽性fociによって占められる肝の面積比などの連鎖解析を行い、化学発がん感受性に関,垢觚彊・篥岨劼寮・・両紊任虜属未魏鮴呂掘▲薀奪搬茖韻・茲啾茖汗色体上に夫々マップされ、さらに詳細に解析中である。
5.臨床への応用、有用性
多段階発がんのイニシエーションよりも後期のプロモーションあるいはプログレッションの段階での発がんを左右、または遅らせることに関する遺伝子の解明に適したモデルラット
