Inbred Dark Agouti (DA) rats were originally obtained from the Zentralinstitut für Versuchstierzucht (Hannover, Germany) and Piebald Virol Glaxo(PVG) rats from Harlan UK Limited (Blackthorn, UK). Animals for congenic breeding were selected from the 10th generation of an advanced intercross line (AIL) originating from the EAE-susceptible DA and EAE-resistant PVG.av1 rat strains. Selected males containing a PVG.av1 fragment in the Eae18a region (from D10Rat81 to D10Rat238 marker, at 49.62 Mb and 62.68 Mb, respectively) were backcrossed with DA females for 10 generations, and offspring were genotyped using microsatellite markers in each generation. In the 10th generation, heterozygous males and females were crossed to obtain the experimental population of homozygous congenic rats and littermate controls. |
