 Origin A transgenic construct was designed with the rat prolactin promoter (-3221 - 3233) controlling EGFP. Transgenic rats originated from Crlj:WI (Wistar) rats. (Jul 16, 2009) 
 Strain Characteristics In adenohypophysial cells, 55% cells are positive for GFP. Among GFP positive cells, 76% are immunostained with prolactin antibody. (Jul 16, 2009) 
 Breeding Conditions The transgene has no effect on fertility. General breeding and reproduction rate. (Jul 16, 2009) 
 Genotyping 1. PCR analysis on genomic DNA. Primer sequences: Forward: 5'-CTGGCCACTATGTCTTCCTGA-3', Reverse: 5'-TGTAGTTGCCGTCGTCCTTGA-3'. Protocols: 95˚C-10 min, 35 cylcles of (94˚C-1 min, 64˚C-1 min, 72˚C-1 min) Product: about 860-bp. 2. Southern blotting with genomic DNA. EGFP + poly(A) (about 1-kb) is detected with EGFP cDNA probe on BamHI-cut genomic DNA. (Jul 16, 2009)
