 Gene Tie2 promoter-GFP (Original plasmid name; pSP14/15.t2hgfpPan5’)
Origin This transgenic strain was generated by microinjection into fertilized oocytes of Wistar rats (Ohtsuki, 2005). The Tek/GFP plasmid (pSP14/15.t2hgfpPan5’) was a given from Dr. TN Sato of University of Texas Southwestern Medical Center at Dallas (Motoike 2000). The transgene was excised from the plasmid vector by Sal I digestion. (Aug 31, 2010)
Strain Characteristics GFP is expressed in capillary endothelial cells under the control of Tek (other name is Tie2) promoter. The blood vessel is visible in this strain. (Aug 31, 2010)
 Breeding Conditions normal breeding
Genotyping Genotyping by PCR analysis.<br />
Extract-N-Amp Tissue PCR kit (SIGMA) Primer 1: 5’-ATTCTCGTGGAACTGGATGG-3’ Primer 2: 5’-GGACAGGTAATGGTTGTCTGG-3’ Signal: 567 bp Cycle Program: 1 cycle of 94C for 3 min, 40 cycles of 94C for 30 s, 55C for 1 min, 72C for 1 min, and a final elongation of 72 C for 2 min. (Aug 31, 2010)
